That is kind of an odd way to describe the fabric I think. Was that off some website someplace? But if it's a knit Polo kind of shirt, like maybe some Golfer's might wear, or work wear with a Company Logo on it, or something that Ralph Lauren might sell with a Pony on the front of it, and buttons or button holes are in the knit part of it, then I think you had better have some interfacing in there, at least under those areas.
Even if it was knit and the collar and stand, and front packet of woven cotton more Oxford cloth, probably would not hurt none to have a bit of interfacing in there either.
I have not actually seen any guys or gals actually wearing Knit Polo kind of shirts, buttoned up all the way at the neck ever though, and I have just always seen them worn more open and unbuttoned instead, at least for the top few buttons, so as long as you were not going to do that, and not even use that top button to keep the shirt closed there, I think it's even more for decoration and would not get a particular lot of stress on that very most top collar button and button hole.
